What is microscopy and what is it used for? There are two major types of electron microscopy. In scanning electron microscopy ( SEM ), a beam of electrons moves back and forth across the surface of a cell or tissue, creating a detailed image of the 3D surface. This type of microscopy was used to take the image of the Salmonella bacteria shown at right, above. A microscope is an instrument that makes an enlarged image of a small object, thus revealing details too small to be seen by the unaided eye. The most familiar kind of microscope is the optical microscope, which uses visible light focused through lenses.

Microscopy techniques are very useful for determining the morphology of particles. The advantages of using microscopy for particle size analysis are that it can be ingredient specific, and is complementary to other particle sizing techniques.
Light microscopy is a general term used for any type of microscopy where light is being transmitted from a source which is on the opposite side of the sample, to the objective lens. Generally, the light is passed through a condenser to focus it on the sample to have maximum brightness. Microscopy was the initial methodology of choice for the study of microorganisms, dating back to the seventeenth century.
